Mechanism

Dantrolene inhibits ryanodine receptor calcium release channels that are expressed on the surface of sarcoplasmic reticulum. Inhibition of these channels causes decreased calcium release and results in skeletal muscle relaxation.

Dose

1 mg/kg IV up to maximum dose of 10 mg/kg

Indications

Malignant hypothermia

Action

Reverses malignant hypothermia by relaxing skeletal muscle

Site of action

Skeletal muscle

Adverse Effects

Dizziness, lightheadedness, and drowsiness most common
